What is Yorvipath used for?

Yorvipath (palopegteriparatide) is commonly used to treat low parathyroid hormone levels, also called hypoparathyroidism.

Yorvipath may also be used for other conditions as determined by your healthcare provider.

How does Yorvipath work (mechanism of action)?

Calcium and phosphate are important minerals in your body. Parathyroid hormone is a natural chemical in your body that balances the levels of calcium and phosphate in your blood. Yorvipath is a medicine used when the parathyroid glands do not produce enough parathyroid hormone to balance calcium and phosphate in the body. It works like your body's parathyroid hormone by keeping hormone levels steady and normal, helping maintain proper calcium and phosphate levels.

How is Yorvipath supplied (dosage forms)?

Yorvipath is available in the following dosage forms that are injected under the skin (subcutaneous).

  • 168 mcg/0.56 mL prefilled pen
  • 294 mcg/0.98 mL prefilled pen
  • 420 mcg/1.4 mL prefilled pen

How should I store Yorvipath?

Before the first use, Yorvipath should be stored in the refrigerator between 36 F to 46 F (2 C to 8 C). After the first time you use it, store it at room temperature below 86 F (30 C). Throw away Yorvipath 14 days after the first use. Store it in the original package with the pen cap on to protect it from light. Do not freeze Yorvipath. Store it away from heat.

Side Effects

What are the most common side effects of Yorvipath?

The most common side effects of Yorvipath are listed below. Tell your healthcare provider if you have any of these side effects that bother you.

  • Pain, itching, swelling, or redness near the injection site
  • Low blood pressure or feeling lightheaded when you stand up (see below)
  • Headache
  • Diarrhea
  • Back pain
  • High calcium level (see below)
  • Sore throat

What are the serious side effects of Yorvipath?

While less common, the most serious side effects of Yorvipath are described below, along with what to do if they happen.

Severe Allergic Reactions. Yorvipath may cause allergic reactions, which can be serious. Stop using Yorvipath and get help right away if you have any of the following symptoms of a serious allergic reaction.

  • Breathing problems or wheezing
  • Racing heart
  • Fever or general ill feeling
  • Swollen lymph nodes
  • Swelling of the face, lips, mouth, tongue, or throat
  • Trouble swallowing or throat tightness
  • Itching, skin rash, or pale red bumps on the skin called hives
  • Nausea or vomiting
  • Dizziness, feeling lightheaded, or fainting
  • Stomach cramps
  • Joint pain

High Calcium Level (Hypercalcemia). Yorvipath can cause high calcium levels, also called hypercalcemia. Hypercalcemia can lead to serious health conditions, such as problems with your kidneys or bones. Hypercalcemia is more likely to happen within the first 3 months of starting Yorvipath, but it can happen at any time. Stop using Yorvipath and call your healthcare provider if you have any of the following symptoms.

  • Peeing frequently
  • Feeling thirsty
  • Tiredness
  • Confusion or dizziness
  • Poor appetite
  • Nausea or vomiting
  • Constipation
  • Muscle weakness
  • Bone pain
  • Abnormal heart rhythm

Low Calcium Level (Hypocalcemia). Yorvipath can cause low calcium levels, which can be serious. Decreased calcium levels can cause a rare dangerous heart rhythm problem called QT prolongation and torsade de pointes. Some people have a higher risk of this, including people who are older, have other people in their family who have had these conditions, have low potassium or magnesium, or who take some medicines for other heart rhythm problems. Your risk of hypocalcemia is higher if you stop using Yorvipath, or if you miss or change your dose, but it can happen at any time. Stop using Yorvipath and call your healthcare provider if you have any of the following symptoms.

  • Abnormal heart rhythm
  • Fainting
  • Tingling in your fingertips, toes, lips, or tongue
  • Numbness in your mouth
  • Muscle pain or spasms
  • Seizures
  • Depression
  • Problems thinking or remembering

Low Blood Pressure Upon Standing (Orthostatic Hypotension). Yorvipath may cause a sudden drop in blood pressure when standing up from a seated or lying position. To help prevent this, be careful not to get up too quickly. If you feel dizzy, sit or lie down right away until the dizziness stops, then take your time getting up again. When you give your first few doses Yorvipath, make sure you are in a place where you can sit or lie down right away if you get these symptoms. Tell your healthcare provider right away if you have any of the following symptoms of orthostatic hypotension. 

  • Dizziness, feeling lightheaded, or fainting upon standing
  • Blurred vision
  • Confusion
  • Feeling weak

Bone Cancer (Osteosarcoma). Yorvipath may increase your risk of a certain type of bone cancer called osteosarcoma. Tell your healthcare provider if you have any of the following symptoms. They may need to do further tests.

  • Pain in any areas of your body that does not go away
  • A lump or swelling under your skin that is tender when you touch it

Who should not use Yorvipath?

Allergies to Ingredients. People who are allergic to any of the following should not use Yorvipath.

  • Yorvipath
  • Palopegteriparatide
  • Any of the ingredients in the specific product dispensed

Your pharmacist can tell you all of the ingredients in Yorvipath.

What should I know about Yorvipath before using it?

Do not use Yorvipath unless it has been prescribed to you by a healthcare provider. Use it as prescribed.

Do not share Yorvipath with other people, even if they have the same condition as you. It may harm them.

Keep Yorvipath out of the reach of children.

Yorvipath is an injection you give under your skin in your abdomen or in the front of your thigh. Before you use Yorvipath for the first time, make sure that you or a caregiver understands how to properly prepare and use it. If you have any questions, talk to your healthcare provider.

Keep all appointments for blood work to check your calcium levels before you start Yorvipath and during treatment.

Always inspect Yorvipath before using it. Yorvipath is a colorless liquid. Do not use it if the liquid is cloudy, colored, or has visible particles in it.

Do not stop using Yorvipath or change your dose unless your healthcare provider tells you to.

What should I tell my healthcare provider before using Yorvipath?

Tell your healthcare provider about all of your health conditions and any prescription or over-the-counter (OTC) medicines, vitamins/minerals, herbal products, and other supplements you are using. This will help them determine if Yorvipath is right for you.

In particular, make sure that you discuss any of the following.

  • Risk for Bone Cancer (Osteosarcoma). Tell your healthcare provider if you have any of the following conditions that may increase your risk of developing bone cancer (osteosarcoma).
  • Bone disease, such as Paget’s disease
  • High levels of an enzyme called alkaline phosphatase
  • Bone cancer or other cancer that has spread to your bones
  • Radiation therapy to your bones in the past
  • Family history of a condition that increases your risk of bone cancer

Other Medicines and Supplements. Yorvipath may interact with other medicines and supplements. Before using Yorvipath, tell your healthcare provider about any prescription or over-the-counter (OTC) medicines, vitamins/minerals, herbal products, and other supplements you are using. See the Interactions section for more details.

Pregnancy. It is not known if or how Yorvipath could affect pregnancy or harm an unborn baby. Tell your healthcare provider if you are or plan to become pregnant. Your healthcare provider will advise you if you should use Yorvipath while you are pregnant or trying to get pregnant. If you become pregnant while using Yorvipath, you can enroll in a Pregnancy Safety Study by calling 844-442-7236. The purpose of this study is to collect information about the health and safety of you and your baby.

Breastfeeding. It is not known if Yorvipath passes into breast milk. Tell your healthcare provider if you are breastfeeding or plan to breastfeed. Your healthcare provider will tell you the best way to feed your baby while you are using Yorvipath.

Does Yorvipath interact with foods or drinks?

There are no known interactions between Yorvipath and foods or drinks.

It is unknown if drinking alcohol will affect Yorvipath.

Does Yorvipath interact with other medicines (drug interactions)?

Yorvipath interacts with many other medicines. Always tell your healthcare provider about any prescription or over-the-counter (OTC) medicines, vitamins/minerals, herbal products, and other supplements you are using.

In particular, make sure that you discuss if you are using any of the following before using Yorvipath.

  • Digoxin (Lanoxin), which is a medicine used to treat irregular heartbeat and some types of heart failure
  • A bisphosphonate, such as alendronate (Binosto, Fosamax), risedronate (Actonel, Atelvia), or ibandronate, which is a medicine commonly used for osteoporosis
  • A diuretic such as hydrochlorothiazide, also called a water pill, which is a medicine used to reduce edema (fluid retention) and blood pressure
  • Lithium, which is a medicine for certain mental health conditions
  • Vitamin A, vitamin D, or calcium supplements
  • Other medicines that can affect calcium levels in your blood

Overdose/Missed Dose

What should I do if I accidentally use too much Yorvipath?

If you or someone else has used too much Yorvipath, get medical help right away, call 911, or contact a Poison Control center at 800-222-1222.

What should I do if I miss a dose of Yorvipath?

If you miss a dose, take it as soon as you remember if less than 12 hours have passed. If you miss your dose by more than 12 hours, skip the missed dose and only take the next dose. Do not take double or extra doses.
